With Tywin dead and Kevan now dead, Jamie wearing white, MIA; who has the best claim to Casterly Rock?
Cersei is gonna blame kevans death on Tyrion in the walls... Lancel?
Does Tyrion have a shot?

No way in hell.  He killed Tywin.  Pretty sure it doesn't work that way.

Tyrion's only hope is getting in good with Dany and her installing him there by force.  Otherwise, he's an accused and convicted kinslayer and kingslayer.  Really, he's a kinslayer twice officially - Joffrey and Tywin.  And even if he can get off the Joffrey charges, it's unlikely that he's going to get out of killing Tywin.

There's also that Lannister who 'went missing' during that riot in KL. Tyrek is pretty high up in the succession line. I think Varys has him, and will use him to claim the Rock.

I really hope none of Tywin's kids get it. Cersei is technically "Lady of the Rock" but she is on trial now and god knows how long it's been since she actually went there. After Tywin's line, it passes to Kevan's son Martyn, skipping over Lancel since I'm sure his vows with the faith bar him from owning land. So yeah, I want Martyn to have it. But Sully is right, Tyrek is very far up the line, and many suspect Varys is keeping him so he can install a pro-Aegon Lannister as lord of the Rock.

Technically the succession goes something like this (feel free to correct me if I made a mistake. Strike is dead, italics is cant inherit for different reasons)

^This. Tyrek shows up again and declares for Aegon, he's nothing but a puppet to Varys.

Lancel joined the faith. If Cersei is killed it goes to Tommen, or Myrcella if he wants to pass it (the Tyrells wouldn't let him though). If they are declared bastards or killed, it goes to Kevan's son Martyn.

Also, Martyn and his sister would be ahead of Tyrek in the line of succession.

If Sansa were cleared of her involvement in Joffrey's death, is there a chance she'd end up inheriting Casterly Rock? There'd be some sweet poetic justice in that.

No.  That's not how it works.  If she had had his son, she might have done what Lysa is doing in the Vale, the same way the Lannisters hoped to control Winterfell by getting Sansa to have Tyrion's son.  But otherwise, her marriage to Tyrion gives her no claim.

I'm not sure about Myrcella and Tommen. Currently, Cersei is set to inherit from Tywin, and if she can't (which is likely because of the trial), it passes to Tywin's next heir, Kevan  Lancel   Willem   Martyn.

I think Varys' plan is to clear the way for Tyrek and then snatch him out of a closet somewhere to be his Westerlands stooge.

Tywin's dead.
Jaime can't inherit.
Tyrion's attainted and, oh yeah, thousands of miles away.
Cersei holds it now but if she dies obviously she loses it.
Tommen can't inherit it because he's the king.
Myrcella would be next, I guess, except she might die too, or become queen and lose it.
Kevan's dead, so move on down to his kids.
Lancel is now a devout who'd turn his back on the whole thing.
Willem is dead.
Martyn is alive still but that could change easily.
Janei is a toddler.
Now on to Tygett, who is dead.
Aaaaand bingo, Tyrek.

Tyrek's (Varys') obstacles to Casterly Rock are a queen on thin ice, a young girl in line for the monarchy, a soldier who has already been taken captive once, and a two-year-old girl.

That's the Iron Throne, where all the males have to be exhausted before a girl can inherit. Normally, boys inherit before their sisters, but girls inherit before their uncles. Unless it's Dorne, where the oldest child inherits, boy or girl.

Girls inherit, but only after boys.  That's why Littlefinger is cultivating Sansa and Roose/Ramsay are doing the whole fake Arya thing.

Wait now I'm confused...the Targ rule of inheritance basically was to look at all male relatives of the current king before looking at the female. Is that also how Westerosi law works?

If it followed the Targ rules then Tyrek would be second in line after Martyn

What if the girl weds?  Does it go to the child they have?  Then a non Lannister can be lord of Casterly Rock?

I'm not sure if Cersei can be Lady of Casterly Rock if she's also the Dowager Queen or the Regent. She's technically a member of House Baratheon of King's Landing, so she would have given up her claim to the Rock by marrying into Robert's family. I don't think she can get it back, even if she is restored to good standing.

The obvious exception would be if she somehow managed to be the only Lannister left alive, but we're pretty far from that scenario.

In that case, Martyn is the next in line.

Couldn't Lancel inherit and then donate the wealth of Casterly Rock to the Faith? The Faith could probably find a lot of uses for all that gold.
