Were anti-magic maesters happy with Rickard's southron ambitions?

Making some pretty big assumptions there both when it comes to Rickard's plants for the future and the Maester.

Buuut let's say that Rickard had nefarious plots below the Neck and the Maesters a) have a Westeros spanning conspiracy and b) work to stamp out magic.

Honestly they seem at odds. Rickard is from a old as snek House with all sorts of weirdo magick that seems defunct.

How trading one bloodline of weirdos with non-working magical powers (The Targs) for another bloodline of weirdos with non-functional magical powers (The Starks) a improvement for the Anti-Mages f the Citadel?

During Aerys reign magic was already dead, off-ing a few Targs a letting the rest scatter to the wind doesn't make much of a difference there.

Putting Robert on the throne didn't really end the Targ line anyway as he is a one fourth Targ.

Magic had just about died at the time.  I doubt the maesters were trying to end magic.  And if they were, well let us say they chose the wrong family to start a campaign against magic.  The Starks used magic in the past to beat the other contenders in the north.  They skin change and give human sacrifices to the Old Gods.

what exactly were Lord Rickards "Ambitions?"

Probably to help feed the North in long winters with food from the fertile Riverlands. Also if Winterfell and Riverrun were joined with marriage the Iron Born would be less likely to target either of them as they would have to deal with both kingdoms. Not a very sexy answer though.
