House Florent and their claim to Highgarden

Florents aren't powerful enough to force their claim.In this, they parallel with the Volmarks.But in their case Hoares were not liked in Iron Islands.

Their window is really closed in main story, with Tyrells being well established and respected.

Did they ever really had any realistic chance of snatching it up after Dragons gave it to Tyrells?

Plenty of chances. Up until the Blackfyre rebellions when Leo Longthorn finally put his foot down, the Tyrells grasp on the Reach was very weak. Between that and the Conquest, both the Florents and the Hightowers could have (and the Hightowers nearly did in the Dance) taken over the Reach.

There were couple loose dragons in Reach when their previous riders were killed during/after Battle of 2nd Tumbleton. So just in theory Florents could have become dragonlords:P

Judging by the strength of such houses as Tarly, Hightower, Redwyne, etc, there was no way that the Florentine could become rulers of the Reach. If GRRM wanted them to be an actual rival to the Tyrells, they should have had more than 2000 swords.

The Hightowers never wanted to take over the Reach. They were just acting on the orders of Aegon II to put down the rebellions in the Reach and later tried to depose Rhaenyra.

By the time of Leo Longthorn Tyrell power should already have been very strong. They were still relatively weak when Jaehaerys I took the throne, but at the end of his reign they established themselves after they got the chance to serve on the council, and later still the Dance would also have played into their hands, weakening many of the great houses of the Reach.
