Why did he do that?

He didn't want Jans Slynt to get Harrenhal because his father was a butcher.

So why did he give the former seat of the second most powerful family of the Westerlands to the grandson of a spice trader?

Because it's a ruin. Just like giving Harrenhal to Janos was ultimately pointless as neither have the finances or the man power to properly restore it.

I don't believe Castamere is a ruin. It might have been damaged in the fighting somewhat, but you guys are thinking of Tarbeck Hall which was brought down on some "scheming bitch."

In defense of Tywin, while the Spicers are a new house, at least the house existed already unlike with Janos. They also have strong ties to the Westerlings who are a very old house. Also Catamere was never "the seat of kings.*" Also these people really helped Tywin win the war. What did Janos do that any other commander of the watch wouldn't do for the money Littlefinger bribed him?

*though what Tywin meant by that is still a little vague, right now we only know of King Harren.

It was something he promised, because Rolph helped in the Red Wedding plans.

Also this. It's made clear at some point that they asked for Castamere when agreeing to the plot.

House Spicer has spice traders origins, but they are a House for a long time

what about House Slynt?

it just popped because he betrayed Ned Stark and helped to get rid of a bastard baby

I agree (as I basically said above) but in truth they haven't been a house for a long time, they're on their third generation I think.

I guess that it was a symbolic gesture.

Reynes lost Castamere and their lives and their House was extinguished when Tywin confronted them.

Since then Castamere was abandoned untill Tywin decided to give it to the Spicers, as a reward for helping Lannisters during the war.

Even though the Spicers are a relatively new House, they are connected to the Westerlings, a rather older House and they have more uses for Tywin in comparison to Janos Slynt.

Slynt was useful in KL, mostly due to the fact that he was a corrupt official. Harrenhal is the largest castle in Westeros and the most notorious and located at Riverlands. Perhaps Tywin didn't trust Slynt or thought that he would take advantage of his sudden rise from Commander of the City Watch to Lord of Harrenhal and expected Slynt to rebel or ask for more.

The Spicers live in the actual place which is a reminder of what Tywin Lannister does to people who move against him.

it's a ruin, but it's the hugest castle in Westeros and it is a military strategic point to defend King's Landing from the North

Why give it to a butcher's son? And we know how Slynt was greedy

Once house Tully was out, they would ask to be Lord Paramounts of the Riverlands, as we see House Baelish is now Paramount. The last thing Tywin (and Tyrion) wanted to see, worms like them as Lord Paramounts

Harrenhal, as a former royal seat, lends tremendous prestige to its holder, elavating him or her to near-Lord Paramount status and therefore among the foremost of noble houses on virtually equal footing with the Great Houses of Westeros. Holding Harrenhal, for example, allowed Littlefinger to marry Lysa Arryn.
