Tyrek was Robert's squire, alongside Lancel, so he may have information about him or Cersei.  That could have resulted in him either deciding to flee on his own initiative, with or without help, or being put on ice by someone, most likely Varys.

If this is what Tyrek's disappearance was about then we won't be seeing him show up later. The war has already been raging and the lines have already been drawn. Are the Tyrells going to give up their hold on the throne just because Tyrek claims Cersei was responsible for Robert's death? Is Stannis going to say he was opposed to the Lannisters before but now he's really opposed? No ones going to care about whatever information Tyrek might have. The other ideas on this thread might be relevant to the rest of the story but Martin isn't going to bring Tyrek back just to talk about Robert and Cersei.

Tyrek being mentioned that often means that GRRM doesn't want us to forget about him. I think he would have gone missing with or without a riot.

Tyrek's into House Hayford made him a target, imo. From what little we know about House Hayford, they have been staunch supporters of House Targaryen and specifically the current branch. One of them was beheaded for telling Maegor to abdicate in favor of Jaehaerys. Another one was beheaded for keeping his oath to Rhaenyra. They sided with the Targaryens over the Blackfyres.

Putting a Lannister as the head of House Hayford doesn't work for someone who is looking to that House to join Aegon's side after he lands.
